The protein encoded by this gene is a member of a family of proteins which inhibit apoptosis through binding to tumor necrosis factor receptor-associated factors TRAF1 and TRAF2. Similar to API1, BIRC4 inhibits apoptosis induced by menadione, a potent inducer of free radicals, and ICE. BIRC4 also inhibits at least two members of the caspase family of cell-death proteases, caspase-3 and caspase-7.

The protein shares no significant homology with IFN-β or the various IFN-α family proteins. IFN-γ was originally characterized based on its antiviral activities. The protein also exerts anti-proliferative, immuno-regulatory and pro-inflammatory activities and is thus important in host defense mechanisms. IFN-γ exerts its biological activities by binding to specific cell surface receptors with high-affinity binding sites.

Cas9 is the nuclease guided by the crRNA and tracrRNA (or trans-activating crRNA) to cleave specific DNA sequences. A guide RNA (gRNA) can be designed to include a hairpin that mimics the tracrRNA-crRNA complex. Binding specificity is based on the gRNA and a three nucleotide NGG sequence called the protospacer adjacent motif (PAM) sequence. With its ease in designing guide sequences to target specific genomic loci, the CRISPR/Cas system is a much simpler, faster, and robust alternative to TALEN and Zinc finger nuclease platforms. For site-specific genome editing, the CRISPR/Cas9 system requires the Cas 9 nuclease and the gRNA. This adenovirus expresses a codon-optimized Cas9 nuclease, along with a mCherry reporter. It can be used along with guided RNA for genome-editing.This adenovirus expresses a FLAG tagged Cas9 with a mCherry reporter.

Syk p72 and ZAP-70 resemble the Src family protein tyrosine kinases in that they have a C-terminal catalytic domain, but differ in that they are characterized by two SH2 domains but no SH3 domains. Syk p72 kinase fused to the transmembrane and extracellular domains of CD7 and CD16, respectively, can induce complete T cell activation, implying the involvement of the Syk/ZAP family proteins in T cell activation. In contrast, the ZAP-70 kinase is insufficient for complete T cell activation unless it is coaggregated with a Fyn p59-containing chimera, suggesting that regulation of ZAP-70 activity may require a functional interaction with Src family kinases.

pCSG-IBA103 is a large expression vector with universal features for transient expression of target proteins with a C-terminal Twin-Strep-tag as well as for generation of stable mammalian cell lines. The vector carries an ampicillin resistance cassette for selection of transformed E. coli cells and ColE1 origin for a high plasmid copy number. Extrachromosomal replication in mammalian cells could occur either by origin of replication from Epstein-Barr Virus (oriP) or by SV40 ori. For the former the vector provides the EBNA-1 gene and for the latter the cell line has to be latently infected with SV40 or express the SV40 large T antigen (e.g. HEK293T, COS-1, COS-7). Stable cell lines can be selected by the neomycin resistance gene (NeoR). In addition, the human cytomegalovirus (CMV) immediate-early promoter enables a high-level expression in a wide range of mammalian cells.

T-cell factor (TCF-1) and lymphoid enhancer-binding factor (for LEF-1, also designated TCF-1(alpha)) compose a family of DNA-binding transcriptional activators that are essential for lymphoid cell development. These transcription factors are activated by the Wnt-1 and Wingless pathways and are characterized by the presence of a conserved protein motif, the high mobility group (HMG) 1 box, which mediates DNA binding. LEF-1, TCF-1, and two other family members, TCF-3 and TCF-4, directly bind to cytosolic beta-catenin and facilitate its translocation to the nucleus, where these complexes induce expression of Wnt target genes.
